人大肠癌细胞株的转移潜能及其相关特性的比较 被引量:8

摘要 目的比较两种人大肠癌细胞株的转移潜能及其相关性.方法利用裸鼠实验性肝转移模型比较人大肠癌HT29与LoVo细胞株的转移潜能;定量检测两种细胞株层粘连蛋白受体(LNR)、上皮性钙调蛋白(Ecad)及Ⅳ型胶原酶的表达;用原位分子杂交及免疫组化检测两细胞株CD44V6在蛋白质水平及mRNA水平的表达.结果LoVo细胞株为人大肠癌高转移株,裸鼠实验性肝转移率为100%;HT29细胞株转移相对较低,裸鼠肝内转移率为429%.定量检测发现,LoVo细胞几乎不表达Ecad,HT29细胞膜上弱表达Ecad.LoVo表达LNR与Ⅳ型胶原酶高于HT29.LoVo细胞内CD44V6的表达在蛋白质水平与mRNA水平均高于HT29细胞.结论LoVo细胞株为人大肠癌高转移株,HT29细胞株转移力相对较低;LNR、Ecad、Ⅳ型胶原酶及CD44V6可作为大肠癌细胞转移潜能的相关指标. IM To compare the metastatic potentials between two human colorectal carcinoma cell lines and to study their features related to metastasis. METHODS Experimental metastatic potentials of two human colorectal carcinoma (HCC) cell lines, HT29 and LoVo, were compared by using nude mice model of HCC liver metastasis. The expressions of Ecadherin, LNR and type Ⅳ collagenase (Ⅳ col) of these HCC cell lines in vitro were observed quantitatively. The cells expressing CD44V6 in mRNA and protein were detected by in vitro hybridization and immunohistochemistry. RESULTS The nude mice had 100% liver metastasis which demonstrated that LoVo is a high metastatic HCC cell line. However, HT29 is a relatively low metastatic HCC cell line since only 429% of nude mice transplanted HT29 cells had liver metastasis. The expressions of Ecad, LNR and Ⅳ col of HT29 and LoVo cells in vitro were detected quantitatively which showed that HT29 cells had weak Ecadherin expression on cell membrane while LoVo cells showed almost no expression. The expressions of LNR and Ⅳ col in LoVo cells both were higher than that in HT29 cells. The expressions of CD44V6 of LoVo cells were higher than that of HT29 cells in either mRNA or protein. CONCLUSION LoVo is a high metastatic HCC cell line, while HT29 is a relatively low metastatic HCC cell line. Ecadherin, LNR, Ⅳ col and CD44V6 can be used to determine the metastatic potential of HCC cells. .
