摘要
目的检测三种不同来源卵巢癌细胞株中67kD层粘连蛋白受体(67kD LN-R)的表达,探讨其在卵巢癌侵袭转移中的作用。方法体外培养腹水来源的人卵巢癌细胞株HRA、SKOV 3及性实体瘤来源的人卵巢腺癌细胞株3AO。以免疫组化和逆转录-聚合酶链反应技术(RT-PCR)检测各细胞株中67kD LN-R蛋白表达及其mRNA水平。结果三种细胞的胞膜及胞浆中67kD LN-R蛋白均呈阳性表达,但HRA、SKOV 3明显强于3AO细胞,差异有显著性(P均<0.05);HRA、SKOV 3均可见明显的474bp的67kD LN-R特异性条带,其mRNA分别为1.156、1.081;3AO特异性条带极弱,其mRNA为0.358;前两者与后者相比,差异有显著性(P均<0.05)。结论67kD LN-R在卵巢癌侵袭转移中发挥重要作用。
Objective: To detect the expression of 67kD laminin receptor (67kD LN-R)in different ovarian carcinoma cells and to study its effect on invasion and metastasis of ovarian carcinoma. Methods. The high malignant obtained from ascites ovarian carcinoma cells HRA,SKOV3 and low malignant carcinoma cell 3AO were cultured in vitro. The expressions of 67kD LN-R at protein level were detected by immunochemistry method, the expressions of 67kD LN-R at mRNA level were detected by semi-quantitive reverse transcript polymerase chain reaction (RT-PCR). Results,The protein expressions of 67kD LN-R in HRA and SKOV3 cell lines were much higher than in 3AO cell lines( P〈0. 05). Specific bands of 474bp which was proved to be the length of 67kD laminin receptor cDNA were found in the HRA,SKOV3 cell lines,the level of 3AO cell lines was far less than the two cell lines( P〈0. 05). Conclusion.The high invasive and established from ascites cell lines HRA and SKOV3 obviously express the 67kD LN-R,which may play a role in the mechanism of the invasion and metastasis of ascites of ovarian carcinoma.
