摘要
目的探索益气化瘀补肾方对慢性脊髓损伤的疗效与作用及机制。方法以BBB功能评分评价大鼠脊髓功能;生化酶标板微孔检测法检测脊髓组织中PLA2的含量;以实时荧光定量RT-PCR方法检测脊髓组织中NGF mRNA、BDNF mRNA的表达。结果脊髓慢性压迫后脊髓组织中bc l2-蛋白表达增加;PLA2含量明显增高;NGF mRNA、BDNF mRNA表达均增强。益气化瘀补肾方治疗后脊髓组织中PLA2含量明显减少;NGF mRNA、BDNF mRNA表达均明显增加。结论益气化瘀补肾方能够减轻脊髓慢性压迫后的功能损害,其机制可能是通过抑制脊髓组织中炎症反应、促进神经营养因子NGF和BDNF的表达而实现。
Objective To investigate the effects and mechanisms of ＂Yiqi Huayu Bushen Decoction＂ in the treatment of chronic spinal cord injury（SCI）. Methods Basso, Beattie and Bresnahan locomotor rating scale was used to assess the functions of spinal cord, ELISA to assay the content of PLA2 in the spinal cord and realtime RT-PCR to detect the expressions of NGF mRNA and BDNF mRNA in the spinal cord. Results The expressions of bcl-2, NGF mRNA and BDNF mRNA, and the contents of PEA2 in the spinal cord were increased in compressive myelopathy rats ; after treatment by ＂Yiqi Huayu Bushen Decoction＂, the content of PLA2 was decreased, but expressions of NGF mRNA and BDNFmRNA were increased. Conclusion ＂Yiqi Huayu Bushen Decoction＂ can relieve chronic spinal compressive injury by suppressing inflammatory reaction and promoting expressions of NGF and BDNF.
