慢性脊髓损伤早期酶学变化与细胞的功能状态 被引量:1

Enzymological changes and functional state of cells at early chronic spinal cord injury

摘要 目的:通过对慢性脊髓损伤早期7种酶的活性变化的观察及一般病理学观察、免疫组化检测,观察在慢性脊髓损伤中酶活性变化与细胞功能状态的关系。方法:实验于1998-11/2000-03在青岛医学院解剖教研室完成。①分组:选用健康家兔16只,随机分为3组:实验组8只,对照组5只,正常组3只。②模型制备:制备慢性脊髓损伤动物模型,对照组(实验对照)动物除不推进活塞外余处理同实验组。正常组(空白对照)家兔不作任何处理。③在慢性压迫性脊髓损伤动物模型的基础上,用组织化学的方法和组织扫描光度测定法对16只家兔脊髓第Ⅴ,Ⅸ板层神经元进行了酶学的测定,用原位末端标记方法及内氏染色观察慢性压迫性脊髓损伤后的一般组织病理学变化及脊髓内神经元和神经胶质细胞功能状态。结果:16只家兔均进入结果分析。①实验组家兔脊髓压迫区第Ⅴ,Ⅸ板层的神经细胞中琥珀酸脱氢酶、乳酸脱氢酶、腺苷三磷酸酶、乙酰胆碱酯酶、单胺氧化酶活性与非压迫区、正常组、对照组同板层的神经细胞中相应的酶活性比较,明显下降(P<0.01),而磷脂酶A2、酸性磷酸酶活性则明显升高(P<0.01)。②内氏染色发现实验组压迫区白质中神经胶质细胞增生。后索较重,侧索及前索较轻;灰质中神经元内氏小体淡染、消失;神经元萎缩,脱失,以Ⅸ板层明显。③原位末端标记阳性细胞为神经胶质细胞,各索内均可见到,后索及侧索明显。灰质中原位末端标记阳性细胞为神经胶质细胞,主要在Ⅰ,Ⅱ板层,数量较少。压迫区相邻段病变次之,远段接近正常。对照组与正常组表现正常。结论:在脊髓慢性受压的过程中,琥珀酸脱氢酶、乳酸脱氢酶、腺苷三磷酸酶、乙酰胆碱酯酶、单胺氧化酶、磷脂酶A2、酸性磷酸酶变化可能是神经元功能退变的重要原因。 AIM: To observe the changes of the activities of 7 enzymes, general pathology and immunohistochemistry at early chronic spinal cord injuries, and investigate the relationship betweenthe change of enzymatic activity and functional status of cells. METHODS: The experiment was carried out in the Department of Anatomy of Qingdao Medical College from November 1998 to March 2000. ①Grouping: Sixteen healthy rabbits were randomly divided into 3 groups: test group (n=8), control group (n=5) and normal group (n=3). ② Model establishment: Animal models of chronic spinal cord injury were prepared, and the rabbits in the control group received the same treatment except pushing piston to compress the spinal cord, and the rabbits in the normal group (blank control) were not given any treatment. ③Enzymatic histochemistry staining, Nissl staining and TUNEL technique were applied to observe the change of the activities of enzymes and general histopathological changes, and functional status of nerve cells and neuroglia cells in the V and IX layers in spinal cord. RESULTS: All the 16 rarebits were involved in the analysis of results.①The succinate dehydrogenase, lactic dehydrogenase, adenosine triphosphatase, acetylcholine esterase and monoamine oxidase in the compressed blocks were obviously decreased as compared with those in non-compressed blocks, normal group and control group of the same layers (P 〈 0.01), however, the activities of phosphatase A2 and acid phospholipas were markedly increased (P 〈 0.01). ② Nissl body of neurons were pale in color or disappeared in gray matter with Nissl staining, and neuron atrophy and declined. Posterior funiculus was obvious, and lateral funiculus, anterior funiculus-was slight about prolifera tion of glial cells mainly in she 9^th layer. ③ The hpoptosis cells were glial cells. Posterior funiculus and ateral funiculus were obvious, and grey matter mainly in the Ⅰ and Ⅱ layers with less quantity. The changes in the adjoining block were obvious. The distal block's changes were almost normal, and those in the control group and normal group were normal. CONCLUSION: The changes of succinate dehydrogenase, lactic dehydrogenase, adenosine triphosphatase, acetylcholine esterase, monoamine oxidase, phospholipase A2 and acid phosphatase may be the important reasons for the degeneration of nerve cell during the process of chronic compression of spinal cord.
