摘要
目的研究大鼠脊髓全横断后神经营养因子-3(neurotrophin-3,NT-3)在运动皮质表达的变化,为进一步探索脊髓损伤后的再生修复机制和神经营养因子治疗脊髓损伤提供实验依据。方法雄性SD大鼠36只随机分为3组:手术组(脊髓横断组),大鼠脊髓在T10-T11节段之间完全横断,按存活时间不同又分为术后1、3、7及14 d组;假手术组,只行椎板切除术;正常对照组。动物到达存活时间点后,应用免疫组织化学ABC法和图像分析技术检测NT-3在运动皮质的表达。结果对照组和手术组运动皮质第V层有NT-3阳性细胞。脊髓全横断后NT-3的表达逐渐增高,在运动皮质于术后3 d达高峰,后缓慢下降。结论脊髓全横断后运动皮质NT-3的表达增加,内源性NT-3的增加可能有利于受损的皮质脊髓束神经元的存活与再生。
Objective Changes of neurotrophin - 3 (NT - 3) expression in rat motocortex following complete spinal cord transection were studied in order to provide the experimental evidences for the mechanisms of regeneration and repair after spinal cord injury and for neurotrophins to treat spinal cord injury. Methods Male Sprague - Dawley rats weighing 240 - 260 g were divided into three groups at random: ①Spinal cord transection group which was randomly subdivided into four subgroups according to the survival time at the day 1, 3, 7 and 14 d after operation (n = 6 for each time point) ,in which the spinal cord of rat was transected completely at thoracic level 10 - 11 ; ②Sham operation group (laminectomy only, n = 6) ; ③Normal control group ( n = 6). The expression of NT - 3 in motocortex was examined by immunohistochemistry and image analysis. Results NT- 3 immunoreactive cells were found in motocortex layer g in both control and spinal cord transection groups. After operation, the expression of NT - 3 increased gradually and reached the peak at the day 3 after operation in motocortex. Conclusion The results suggest that the expression of NT- 3 increases in motocortex after spinal cord injury and that the increase of endogenous NT - 3 may be: beneficial for the survival and regeneration of the injured corticospinal neurons.
