摘要
目的:观察乌芪通络胶囊对糖尿病周围神经病变大鼠坐骨神经组织中神经生长因子(NGF)和胰岛素样生长因子-1(IGF-1)的mRNA表达的影响,初步探讨乌芪通络胶囊治疗糖尿病周围神经病变的机制。方法:雄性SD大鼠80只,造模前随机选择14只为正常对照组,造模组66只。后者经腹腔注射链脲佐菌素(STZ)造模,造模成功51只。将模型组随机分为弥可保组、乌芪通络胶囊组、模型组,每组17只。连续给药8周处死,取坐骨神经,抽提总RNA,采用实时荧光定量PCR法检测坐骨神经组织中NGFmRNA、IGF-1mRNA的表达水平。结果:模型组NGF mRNA及IGF-1 mRNA的表达比正常对照组显著降低(P<0.05),乌芪通络胶囊组和弥可保组NGF mRNA及IGF-1 mRNA的表达与模型组相比显著升高,差异有统计学意义(P<0.05)。结论:NGFmRNA和IGF-1 mRNA在糖尿病大鼠坐骨神经中表达降低,乌芪通络胶囊可促使糖尿病大鼠坐骨神经中的NGFmRNA和IGF-1mRNA表达增加,这可能是乌芪通络胶囊治疗糖尿病周围神经病变的机制之一。
Objective：To explore the mechanism of Wuqi Tongluo capsule on DPN,we observed the mRNA expression of NGF and IGF-1 in sciatic nerve of DPN rats.Methods：80 male SD rats were randomly divided into two groups： blank group（14 rats）and model group（66 rats）.The latter was received STZ Intraperitoneal injection with the success of 51 models.The DPN model group was randomly divided into Methycobal group,Wuqi Tongluo capsule group,model group,with 17 rats in each group.Continuous administration for eight weeks,and they were extracted total RNA from sciatic nerve.The NGFmRNA and IGF-1mRNA expression level in sciatic nerve of each group was determined by reverse transcriptase and relative quantity PCR.Results：The expression of NGF mRNA and IGF-1 mRNA in model group decreased more significantly than the blank group（P0.05）.Wuqi Tongluo capsule group and Methycobal group significantly increased compared with the model group,and the difference was statistically significant（P0.05）.Conclusion：NGF and IGF-1 can reduce mRNA expression in sciatic nerve of DPN rats.Wuqi Tongluo capsule can increase the expression of NGF mRNA and IGF-1 mRNA in DPN rats,which may be one of the mechanisms of Wuqi Tongluo capsule in the treatment of diabetic peripheral neuropathy.
