摘要
为了探索鸡白细胞介素-18(IL-18)在鸡球虫病疫苗免疫中的免疫佐剂作用,用鸡IL-18的重组真核表达质粒pIL-l8、柔嫩艾美耳球虫早熟株疫苗及两者联合分别接种3日龄鸡,首免后第1 d、4 d、6 d、9 d、14 d、21 d、26d、28 d时随机抽取鸡外周血及无菌采取鸡脾脏,应用ELISA和甲基噻唑基四唑(MTT)法分别检测免疫鸡的体液免疫及细胞免疫水平。二免后第14 d用柔嫩艾美耳球虫进行攻击。结果表明,pIL-18与鸡柔嫩艾美耳球虫早熟株疫苗联合免疫具有明显提高虫苗免疫保护的作用,pIL-18单独注射鸡体也具有明显增强鸡体抗球虫感染的作用。表明鸡IL-18能够明显增强鸡球虫病疫苗的免疫原性,显著提高免疫鸡的体液免疫和细胞免疫水平,并且能强有力地抵抗柔嫩艾美耳球虫的攻击。
3-day-old SPF chickens were inoculated intramuscularly with recombinant plasmid pIL-18, pIL-18 and coceidiosis vaccine or coccidiosis vaccine. Chicken spleen lymphocytes were detected by methyl thiazolyl tetrazolium(MTT) and the sera were coUected to examine antibodies of chickens by ELISA on 1 d ,4 d ,6 d ,9 d, 14 d ,21 d ,26 d and 28 d after first immunization. The chickens were challenged on day 14 post the booster. The results showed that chicken IL-18 was able to improve obviously the immunogenicity of coccidiosis vaccine, and enhance the humoral immunity level and cellular immunity level of the vaccinated chickens. Therefore,chickens vaccinated with pIL-18 and coccidiosis vaccine were protected effectively against challenge with eimeria coccidia.
