浓香型大曲微生物群落结构的PCR-SSCP分析条件优化 被引量:8

Optimal Conditions of PCR-SSCP Technology for the Analysis of Microbial Communities in Luzhou-flavor Daqu

摘要 文章研究了浓香型大曲微生物群落结构的PCR-SSCP分析条件的优化,结果表明:PCR产物100℃变性5 min后,在交联度为49:1,浓度为8%的聚丙烯酰胺凝胶中,无甘油,4℃条件下200 V电泳15 h,可得到较理想的图谱。 The optimal conditions of PCR-SSCP Technology for the analysis of microbial communities in Luzhou-flavor Daqu is studied in the paper. The results show that under the condition of the concentration of the gel being 8%, the proportion of acrylamide and N, N'-methylene-bisacrylamidei being 49: 1, without glycerine, denaturing at 100℃ for 5 min, 4℃, 200 V, electrophoresis 15 h, perfect spectrogram can be achieved.
