摘要
目的:探讨和尝试一种较为迅速确定最佳退火温度的方法来扩增目的基因。方法:采用梯度PCR对醛固酮合酶基因(CYP11B2)进行扩增,通过凝胶呈像技术判断CYP11B2基因最佳退火温度。结果:与普通PCR相比,梯度PCR能够迅速地确定CYP11B2基因的最佳退火温度。结论:梯度PCR简单、直观、快速,适用于各种核酸分子的扩增,具有高效率、高通量的优点,有着广阔的应用前景。
Objective:To explore a method for quicker identification of the optimal annealing temperature in PCR. Methods :Gradient PCR was employed in analysis of aldosterone synthase (CYPllB2)gene polymorphisms ,and gel Imaging Technology was used to determine the optimal annealing temperature. Result:Compared with normal PCR,Gradient PCR could identify the the optimal annealing temperature of aldosterone synthase gene. Conclusions:Gradient PCR is simple,efficient and rapid,with a broad application prospects, which suitable for PCR of all kinds of genes.
