陇东绒山羊微卫星多态性及其与部分经济性状相关性的研究 被引量:4

摘要 为了在陇东绒山羊群体实施标记辅助选择,根据山羊遗传连锁图谱和相关文献资料,选用9个微卫星座位,8%非变性聚丙烯酰胺凝胶电泳PCR扩增产物,对我国特有遗传资源50只陇东绒山羊基因组DNA的遗传多样性进行了检测,通过标记多态性与生产性状的最小二乘线性模型,进行了分子标记与经济性状的相关分析。结果表明:9个微卫星座位在陇东绒山羊群体中共检测到90个等位基因,平均每个座位的等位基因数为10个。其中微卫星座位LSCV24等位基因数最多,为14个,等位基因数最少的是DB6,只有6个;在所标记群体内平均多态信息含量为0.84±0.03,平均遗传杂合度为0.86±0.03。在9个基因座上发现与某些经济性状密切相关的标记效应(P<0.05),同时在这些位点上找到各经济性状优势基因型以及对相应性状具有正效应或负效应的等位基因。 Nine microsatellite loci selected according to the genetic map of male goat and related reports were studied to reveal genetic diversity of the population including 50 Longdong cashmere goats, a special chinese genetic resource,by 8% nondenatured polyacrylamide gel electrophoresis detecting the product of PCR for MAS. The relating effects between markers and economic traits were analyzed in a generalized linear mode.The results indicated that the total number of alleles were 90, average allele numbers were 10. allele numbers of LSCV24 were 14 while allele numbers of DB6 were 6. The average PIC values and the average genetic heterozygosity were (0.84±0.03) and (0.86±0.03)in Longdong Cashmere goats ,respectively. Nine microsatellite loci had significant effect on some economic traits of Longdong Cashmere goats ,meanwhile, we also discovered that favorable genotype and some alleles above loci had positive effect and negative effect on interrelated economic traits .
