摘要
KAP6.1基因属于角蛋白家族,羊毛纤维的复杂结构主要由角蛋白组成,这些蛋白对于羊毛纤维的机械性能和主要构造起主要作用。本研究以693只中国美利奴羊(新疆军垦型)为实验材料,采用PCR技术、SSCP技术和克隆测序的方法分析KAP6.1基因的多态性。结果表明:KAP6.1基因存在C159T碱基突变和57 bp的插入/缺失,其中,C159T位点存在3种基因型,分别为AA,BB和AB型,基因型频率依次为0.3694,0.4574,0.1732,等位基因A频率为0.4560,等位基因B频率为0.5440。57 bp的插入/缺失位点存在2种基因型AA型(303 bp)和AB型(303 bp/246 bp),基因型型频率依次为0.9292和0.0707,等位基因A频率为0.9646,等位基因B频率为0.0354,实验证明KAP6.1基因能够作为优质细毛羊毛性状的候选基因进行研究。
KAP6.1gene belongs to keratin-associated proteins(KAP)family,the wool fibre is a complex structure composed primarily of proteins from KAP gene family.These proteins are responsible for the major structural and mechanical properties of the wool fibre.PCR,SSCP and sequencing analysis were used to identify polymorphisms of KAP6.1gene in Chinese Merino(Xinjiang Junken Type).The results showed KAP6.1gene had C159T mutant site and 57bp insert/delete site.Three genotypes of C159T site existed in Chinese Merino(Xinjiang Junken Type).AA genotype frequency was 0.3694,AB genotype frequency was 0.1732,BB genotype frequency was 0.4574.A allele frequency was 0.4560,B allele frequency was 0.5440.Two genotypes of 57bp insert/delete site existed in Chinese Merino(Xinjiang Junken Type).AA(303 bp)genotype frequency was 0.9292,AB(303bp/246bp)genotype frequency was 0.0707.A allele frequency was 0.9646,B allele frequency was 0.0354.The results showed KAP6.1 gene could be candidate gene to study wool quality of merino sheep.
