摘要
目的观察“七叶灵”颗粒对人肺腺癌H460细胞生长的抑制作用，以及对H460细胞中COX-2、OPN和hTERT表达的影响，探讨“七叶灵”颗粒在抑制肿瘤细胞生长、延缓肿瘤细胞的转移和浸润方面的作用靶点。方法MTT法测定“七叶灵”颗粒对人肺腺癌H460细胞生长的抑制率，根据结果得出IC50值。用RT-PCR法检测不同浓度的“七叶灵”颗粒对COX-2、OPN和hTERT表达的影响。结果①“七叶灵”颗粒对人肺腺癌H460细胞的生长有抑制作用。②IC50值随时间的增加明显降低（P〈0．05）。③RT-PCR法结果显示：“七叶灵”颗粒抑制了COX-2、OPN和hTERT表达（P〈0．05）。结论“七叶灵”颗粒具有益气养精、清热解毒、消肿散结的功效，有抑制人肺腺癌H460细胞生长的作用，并通过调节COX-2、OPN和hTERT的表达，起到抑制肿瘤细胞生长，延缓肿瘤细胞转移和浸润的作用。
Objective： To observe the inhibition （suppressing） effect of ＂Qi-ye-ling＂ herbal granule human lung adenocarcinoma H460 cell in vitro, and the impact of it on H460 cell COX-2, OPN and hTERT expression.. To discuss the effect target of herbal granule ＂Qi-ye-ling＂ in inhibiting （suppressing） the growth, transfer and infiltration of tumor cells. Methods： MTT method was used to investigate the inhibiting rate of. ＂Qi-ye-ling＂ herbal granule on human lung adenocarcinoma H460 cell, the IC50 value can be inferred from the result. The expression of COX-2, OPN and hTERT was detected by RT-PCR method confirms the affection of ＂Qi-ye-ling＂ herbal granule in different concentration. Results： （ 1 ） ＂Qi-ye-ling＂ herbal granule can inhibit human tung adenocarcinoma H460 cell. （2） As timg going on the IC 50 value decreased obviously （P 〈 0.05 ）. （ 3 ） RT-PCR results showed that the expression of COX-2, OPN and hTERT was inhibited by ＂Qi-yc-ling＂ herbal granule （P〈0.05） . Concusions： ＂Qi-ye-ling＂ herbal granule is clinically proved to be effective in notifying Qi and essence, clearing anay heat and toxin and dissipating snollen and lung in terms of Traditional Chinese Medicine. It can inhibit human lung adenocarcinoma H460 cell in vitro, and can inhibit the growth, transfer and infiltration of tumor cells by regulating the expression of COX-2, OPN and hTERT.
