摘要
目的探讨低分子肝素在兔小梁切除术中抗瘢痕的作用。方法将24只(48只眼)新西兰大白兔随机分为3组:A组、B组、C组,每组8只兔,各16只眼。3组双眼均采用常规小梁切除术,A组术毕和术后第1天结膜下注射0.5 mL平衡盐溶液,B组和C组分别注射低分子肝素10、50 U/mL。3组于术后第3、7、14、28天分别随机选取2只兔处死,取滤过泡区结膜和巩膜组织行PCNA免疫组化染色观察3组术后第3、14天结膜下滤过区成纤维细胞增殖情况,Masson染色观察3组术后第28天结膜下滤过区胶原纤维生成情况。结果术后第3、14天C组结膜下滤过区成纤维细胞PCNA阳性细胞明显少于A组和B组(P<0.05),A组、B组新生胶原纤维量与C组比较差异均有统计学意义(均P<0.05)。结论低分子肝素可抑制成纤维细胞增殖和胶原纤维生成。
Objective TO explore anti-scaring effect of low molecular weight heparin after trabeculectomy in rabbit. Methods 48 eyes of 24 new zealand white rabbits were randomly divided into three groups:group A,group 13 and group C,16 eyes of 8 rabbits in each group. Both eyes in three groups were treated with routine trabeculectomy. Subconjunctival injections of one of three materials were made:0.5 mL balanced salt solution in group A, 10 U/mL low molecular weight heparin (0.5 mL) in 13 group,50 U/mL low molecular weight heparin (0.5 mL) in group C at the end of operation and 1 day postoperatively. Two rabbits selected randomly in each group were killed on days 3,7,14,28 day postoperatively. Newborn collagen fibers of subconjunctival filtering area on 28 th day through masson staining and proliferation of fibroblast cell of subconjunctival filtering area on days 3,7, 14, 28 day postoperatively were observed by immunohistochemistry staining of proliferative cell nuclear antigen(PCNA) in conjunctiva and sclera of filtering area of the rabbits. Results There was significant difference in newborn collagen among group A,group B and group C(P〈0. 05). On day 3,14 postoperatively, the numbers of pcna positive cells in group C were less than those in group A and group B (P〈0.05 respectively). Conclusion Low molecular weight heparin can inhibit the proliferation of fibroblast and collagen fiber.
