摘要
对5-羟色胺（5-HT）抗独特型单克隆抗体的功能进行鉴定．方法：细胞培养法，免疫组织化学ABC法和形态计量法．结果：培养平滑肌细胞显示5-HT受体免疫反应阳性，在正常培养基培养的平滑肌细胞的平均长度为（18．8±35．4）μm．培养基中含不同浓度的5-HT抗独特型单克隆抗体（分别为0．01％，0．03％，0．10％，0．30％，1％抗体原液），平滑肌细胞的平均长度分别缩短为（175．6±43．3）μm，（16．2±46．2）μm，（116．9±25．8）μm，（89．5±22．8）μm，（79．9±21．6）μm，除含0．01％抗体原液组外均比正常组有非常显著的差别（P＜0．01）．结论：该抗独特型单克隆抗体具有与5-HT相类似的诱导平滑肌收缩的功能，5-HT受体可能介导了这种功能．
Aim:To identify the function of 5-HT anti-idiotypic monoclonal antibody. Methods: The cell culture immunohistochemistry and morphometry were used. Results:The cultured smooth muscle cells showed 5-HT receptorimmunoreactivity. The length of smooth cells in normalmedium was (187.±35. 4)μm. It became (175. 6± 43. 3)μm, (160. 2±46. 2)μm, (116. 9± 25. 8)μm, (89. 5±22. 8 )μm, (79. 9± 21. 6 )μm, in medium containing0. 01 %, 0. 03%, 0. 1%, 0. 3%, 1% 5-HT anti-idiotypicmonoclonal antibody respectively, and was significantlyshorter than in normal medium respectively (P<0. 01 ) except of the group in medium containing 0. 01% 5-HT antiidiotypic monoclonal antibody. Conclusion: 5-HT anti-idiotypic monoclonal antibody, similar to 5-HT, possesses afunction inducing smooth muscle contraction and 5-HT receptor may mediate the function.
