雄激素受体、凝血酶敏感蛋白-1与前列腺良、恶性疾病血管生成的关系 被引量:1

The relationship between androgen receptor,thrombospondin-1 and angiogenesis in nonmalignant and malignant prostate tissue

摘要 目的:探讨雄激素受体(Androgen receptor,AR)、凝血酶敏感蛋白-1(Thrombosopndin-1,TSP-1)在良性前列腺增生(Benign prostatic hyperplasia,BPH)、前列腺癌(Prostate cancer,Pca)中的表达及与血管生成的关系。方法:良性前列腺增生20例;前列腺癌60例,前列腺癌组中根据Gleason分级,Gleason评分<7为低评分组前列腺癌(35例),Gleason评分≥7为高评分组前列腺癌(25例);无骨转移者47例,有骨转移者13例。运用免疫组化二步法检测各组中AR、TSP-1的表达水平及微血管密度(Microvesel density,MVD)。结果:BPH组中AR、TSP-1的表达高于Pca组(P<0.01),MVD值低于Pca组(P<0.01);在Pca中低评分组AR、TSP-1的表达高于高评分组(P<0.01),MVD值低于高评分组(P<0.01);无骨转移组AR、TSP-1的表达高于骨转移组(P<0.01),MVD值低于有远处转移组(P<0.01)。AR、TSP-1阳性组的MVD值分别小于阴性组的MVD值(P<0.01)。TSP-1与AR呈正相关(r=0.722,P<0.05)。结论:AR、TSP-1在前列腺癌中均为低表达,且与肿瘤的Gleason评分及有无骨转移密切相关。前列腺癌中AR突变使TSP-1减少,从而促进血管生成。 Objective: To investigate the expression of androgen receptor(AR) and thrombospondin-1 (TSP-1) in human benign prostatic hyperplasia (BPH)and prostate cancer(Pca), and to explore the relationship between them and angiogenesis. Methods: Eighty specimens from BPH (20)and Pca( 60 )were included in the study.The Pca was divided into low Gleason score group (Gleason score〈7 ) and high Gleason score group(Gleason score ≥ 7 ), and into non-bone metastasis group and bone metastasis group. The expression of AR, TSP-1 and microvesel density( MVD ) were detected by immunohisto- chemistry (two-step method ). Results: The positive rates of AR and TSP-1 in BPH group were significantly higher than those in Pca group;the MVD mounts in BPH group was significantly lower than that in Pca group.The positive rates of AR and TSP-1 in lower Gleason score group were significantly higher than those in higher Gleason score group, and the MVD mounts in lower Gleason score group was significantly lower than that in higher Gleason score group.The positive rates of AR and TSP-1 in non-bone metastasis group were significantly higher than those in bone metastasis group, and the MVD mounts in non-bone metastasis group was significantly lower than that in bone metastasis group.MVD mounts in AR positive and TSP-1 positive group were significantly lower than those in AR negative and TSP-Inegative group(all P〈0.01 ).There was positive correlation between the expression of AR and TSP-1 (r=0.722,P〈0.05 ). Conclusion: Low expressions of AR and TSP-1 in Pca have close relations with tumor Gleason score and condition of bone metastasis.Mutation of AR might repress TSP-1 ,which might promote angiogenesis in prostate cancer
