摘要
目的探讨垂体瘤中凝血酶敏感蛋白-1(TSP-1)、CD36、转化生长因子β1(TGF—β1)的表达及其与肿瘤侵袭性的关系。方法取52例垂体瘤(侵袭性组24例,非侵袭性组28例)手术标本,采用RT-PCR检测TSP-1、CD36、TGF-β1的mRNA表达,Western blot方法检测TSP-1、TGF-β1蛋白表达,并分析其表达与垂体瘤侵袭性等因素之间的关系。结果与非侵袭组相比,侵袭组TSP-1 mRNA及蛋白表达较弱,TGF-β1 mRNA及蛋白表达则强于非侵袭性组(P〈0.05),两组间CD36 mRNA表达差异无显著性意义(P〉0.05)。肿瘤的大小组间、各内分泌功能分型组间TSP-1、TGF-β1、CD36差异均无显著性意义(P〉0.05)。结论TSP-1、TGF—β1 mRNA及蛋白表达与垂体瘤侵袭性密切相关,提示其可能在垂体瘤发生发展中起重要作用,并能影响垂体瘤的侵袭性。
Objective To explore the expressions ofthrombospondin-1 (TSP-1), transforming growth factor β1 (TGF-β1), CD36 in pituitary adenomas and investigate the relationships between the expressions and the invasiveness of adenomas. Methods The mRNA expressions of TSP-1, TGF-β1, CD36 in 52 surgical specimens (24 invasive cases; 28 noinvasive cases) of pituitary adenomas was measured using RT-PCR and Western blot method was used to detect the protein levels of TSP-1 and TGF-β1. The correlations between the expressions and invasiveness of adenomas were analyzed. Results In invasive group as compared with the noinvasive group, the expression of TSP-1 (mRNA: t=4.389, P=000; protein: t=4.207, P=0001) was lower, and the expression of TGF-β1 (mRNA: r= 3.356, P=004; protein: t=5.59, P〈0.0001) was stronger, and the mRNA expression of CD36 (t=2.279, P= 0.063) was not significantly different. No difference in the expressions of TSP-1, TGF-β1 and CD36 was seen between different groups of various tumor sizes and endocrine functional types. Conclusion TSP-1 and TGF-β1 maybe play an important role in the development of pituitary adenomas and regulate its invasiveness.
