摘要
利用壳多糖缓慢降解的特性,把它与阿霉素制成壳多糖阿霉素缓释药粒,植入家兔胫骨上端进行体内释放试验,并与静注相同剂量阿霉素的家兔进行比较。结果表明,该缓释药可使阿霉素在局部骨组织中维持较高浓度达8周以上,且骨组织中最高浓度是静注相同剂量阿霉素时骨含量的29倍,但血浆峰值浓度仅为静注组血浆峰值的1/13。因此,壳多糖与阿霉素制成的缓释药粒具有良好的缓释功能,该缓释药粒阿霉素的一次局部用量可超过静注一次用量的10余倍,能较好地满足骨肿瘤局部化疗用药量大、维持时间长、血液浓度低、全身反应小的用药要求。
Chitin is a non toxic and side-effects biological material with good biological compatibility and biodegradation. We used its characteristics to make out adriamycin(ADM) loaded chitin drug delivery system(DDS).The DDS was inserted into intercondylar region of tibia in rabbits and the in vivo release test was done, and compared to that of rabbits with the same dose of ADM injected by vein. The results showed that high concentration of ADM could exist for 8 weeks in local bone of rabbits implanted with the DDS. The maximum content of ADM in bone of rabbits implanted with DDS was 29 times higher than that of rabbits injected the same dose ADM by vein, while the maximum content of ADM in plasma of rabbits with DDS was only 1/13 of that in rabbits with the same dose injected by vein. It is concluded that ADM loaded chitin drug delivery system had good sustained release function, and one dose of ADM with DDS for local use could be 10 times more than that of ADM used in vein injection. So the DDS could meet the needs (large dose, long time sustaining, low blood content and little general reaction) of local chemotherapy for bone tumor.
