摘要
目的:扩增并测定陕西地区丙型肝炎病毒(HCV) 的基因组5’非编码区(NCR)的序列.方法:应用RT-PCR法和基因重组技术将从陕西省NANB病毒性肝炎患者血清中扩增出的5’NCR长262bp的片段,用全自动序列分析仪测序.结果:所得序列与HCV-J6,HCV-J8,HCV-Hebei, HCV-1及HCV-J比较,其同源性分别为99.2%,96.5%,94.6%,94.6%及94.3%,与HCV-J6的同源性最高.结论: 所分离HCV与HCV-J6同属Ⅲ型,
To clone and sequence 5'NCR cDNA of hepatitis C virus(HCV) derived from a patient with HCV infection in Shaanxi. Methods: A 262 bp fragment in 5'NCR was amplified by reverse transcription polymerase chain reaction (RT-PCR) , its sequence was determined by auto-sequencing instrument and was compared with those of the corresponding regions of HCV-J6,HCV-J8,HCV-Hebei,HCV-1 and HCV-J genomes. Results: The nucleotide sequence homologied to those strains were found to be 99. 2%, 96. 5%,94. 6%,94. 6% and 94. 3% respectively. Conclusion: The results indicates that this isolated HCV strain is close to HCV-J 6 and belong to type Ⅲ.
