纤维素酶产生菌黑曲霉的选育及其产酶条件的研究 被引量:10

摘要 进行优良纤维素酶高产菌株的选育,并研究其发酵条件。以黑曲霉菌株S1为原始出发菌株进行紫外诱变,经过单因素实验和正交实验优化突变菌株的产酶条件。获得一株高产纤维素酶黑曲霉(Aspergillus niger)菌株S12,其最佳产酶条件为:在pH6.0、培养温度28℃、培养时间96h条件下;秸秆粉2.5g、麦麸2.5g、1%(NH4)2SO410mL。在此条件下,滤纸酶活力为3.79IU/mL,羧甲基纤维素酶活力19.06IU/mL,β-葡萄糖苷酶活力47.33IU/mL,其酶活分别是原始菌株的1.20倍、1.70倍和1.13倍。 The breeding of high-cellulase-yield strains was operated and the fermentation conditions of the selected strains were studied. As.niger strain S 1 was used as mutan strain for ultraviolet irradiation and the cellulase-producing conditions of the mutant strain were optimized through single-factor test and orthogonal test. Finally a high-cellulase-yield strain As. niger S12 was obtained and its optimum eellulase-producing conditions were summed up as follows: pH value was 6.0, culture temperature was at 28℃ and 96 h culture time, the use level of stalk meal and wheat bran and 1% (NH4)2SO4 were 2.5 g, 2.5 g and 10 mL respectively. Under the above conditions, its enzyme activities toward filter paper (FPA), CMCase and β-glucosidase reached 3.79 IU/mL, 19.06 IU/g and 47.33 IU/mL respectively (increased by 1.20, 1.70 and 1.13 times, respectively).
