蛋白质免疫印迹技术检测肺癌组织上皮钙粘蛋白的表达 被引量:8

摘要 目的探讨上皮钙粘蛋白(E-cad)与肺癌及其病理类型之间的相关性。方法应用蛋白质免疫印迹技术(W estern b lot)检测30例肺癌患者的癌、癌旁和远癌肺组织及5例肺良性对照标本中E-cad的表达,并对部分表达阴性的标本用免疫组化方法进行验证。结果癌组织、癌旁组织、远癌组织E-cad的阳性率分别为36.7%、70.0%、96.7%。癌组织E-cad阳性率明显低于癌旁组织(P<0.017),也明显低于远癌组织(P<0.017);癌旁组织E-cad阳性率明显低于远癌组织(P<0.017)。在30例肺癌患者鳞癌、腺癌及腺鳞癌的癌组织中E-cad阳性率分别为33.3%、40.0%、33.3%;鳞癌、腺癌及腺鳞癌的癌旁组织中的阳性率分别66.7%、73.3%、66.7%;鳞癌、腺癌及腺鳞癌的远癌组织中的阳性率分别为91.7%、100.0%、100.0%。癌、癌旁和远癌组织中E-cad的表达与肺癌组织学类型无关(P>0.05)。结论E-cad蛋白表达减少或缺失与肺癌有相关性,与肺癌的组织类型无相关性。 Objective To explore the relationship between E-cadherin(E-cad) and lung cancer and its pathological types. Methods The expression of E-cad protein in 30 cases of lung cancer tissues, paracancer lung tissues, farcancer lung tissues and 5 cases of non-cancer lung tissues were detected by Western blot analysis. Some negative results of Western blot analysis were confirmed by the immunobistochemical method. Results The positive expression rate of E-cad was 36.7% in lung cancer tissue,70.0% in paracancer tissue and 96.7% in far-cancer lung tissue. There was a significant difference between lung cancer tissue and paracancer tissue or far-cancer tissue ( P 〈 0.017 ). There was also a significant difference between paracancer lung tissue and far-cancer lung tissue (P 〈 0. 017). In the 30 cases of lung cancer tissue, the positive expression rate of E-cad in different histology type was 33.3% in squamous cell carcinoma (SCC) , 40.0% in adenocarcinoma(AD) and 33.3% in adenosquamous carcinoma(AdCa). In the 30 cases of paracancer lung tissue, the positive expression rate was 66.7% in SCC, 73.3% in AD and 66.7% in AdCa. In the 30 cases of far-cancer lung tissue, the positive expression rate was 91.7% in SCC, 100.0% in AD and 100.0% in AdCa. The expression of E-cad in cancer, paracancer and far-cancer lung tissues had no dependability with the pathological type of lung cancer ( P 〉 0.05 ). Conclusions Under regulation or absence of E-cad protein expression would have correlation with lung cancer, but no evident correlation with different pathological types of lung cancer.
