微小RNA-200c上调E-钙黏蛋白表达对外阴鳞癌A-431细胞侵袭及迁移能力的影响 被引量:1

摘要 微小RNA(microRNA,miRNA)是一类由长约22个(18~25个不等)核苷酸组成的不编码蛋白质的RNA,通过抑制靶mRNA的翻译或降解靶mRNA来调节基因的功能,从而参与一系列的重要进程.研究表明,miRNA表达异常与多种肿瘤相关,其可以起到肿瘤抑制基因或者癌基因的功能,其功能是在转录后(翻译水平)负调控基因的表达.miRNA对mRNA异常的转录后调节可以促进细胞增殖、抑制细胞凋亡及增强细胞的侵袭能力,从而导致肿瘤的发生和发展.其中,miRNA-200c(miR-200c)与上皮源性肿瘤的发生、发展关系密切.本研究利用基因转染技术将miR-200c转染至外阴鳞癌A-431细胞,通过检测A-431细胞转染前后E-钙黏蛋白(E- cadherin,E-cad)的表达情况及细胞侵袭能力的变化,分析miR-200c对外阴鳞癌A-431细胞侵袭、迁移能力的影响,并初步揭示其可能的机制.
