水芹总酚酸对小鼠CCl_4肝损伤的保护作用 被引量:11

摘要 目的观察水芹总酚酸(OJTPA)对四氯化碳(CCl4)致小鼠急性肝损伤的保护作用。方法将180只小鼠随机分为正常组,联苯双酯阳性药物组,CCl4模型对照组和OJTPA高、中、低(500、250、125mg·kg-1)3个剂量组。应用0.1%CCl4腹腔注射致小鼠急性肝损伤模型,通过测定小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性以及小鼠肝组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力,并观察肝脏组织病理学变化,对OJTPA的保肝作用进行研究。结果模型组小鼠肝功能明显异常,肝组织变性、坏死严重;OJTPA组能明显改善CCl4中毒小鼠肝功能,其降酶率(ALT)以高、中、低剂量分别为74.74%、55.87%、7.25%;肝组织切片镜检证实,OJTPA能减轻中毒小鼠肝细胞损伤程度。结论OJTPA对急性肝损伤小鼠有较好的保肝作用。 Aim To observe the protective effect of total phenolics acid of Oenanthe javanica (OJTPA) on mice models of CCl4 acute liver injury. Methods 180 mice were randomly divided into the normal group, bifendate control group, CC14 model group and three OJTPA groups[ high (500mg.kg^-1), middle (250mg.kg^-1) and low (125mg. kg^-1 ) dosage group Ⅰ in the experiment. Acute hepatic injury models of mice were induced by 0.1% CCl4 through intraperitoneal injection. The protective liver effect of OJTPA was observed by measuring ALT, AST activity in serum and MDA content, SOD activity in hepatic tissue. Liver histopathological changes were also observed. Results In the model group, the level of ALT,AST increased significantly,and hepatic tissue degeneration was obviously seen. In OJTPA groups , the level of ALT dropped significantly( the restraint rate of the high,middle and low dosage group was 74.74% ,55.87% ,7.25% respectively). Liver histological section test under a microscope proved that OJTPA could relieve the degree of hepatic injury of mice induced by CCl4. Conclusion OJTPA has good protective effect on mice with acute liver injury.
