摘要
目的 观察水芹提取物在乙型肝炎病毒 (HBV)基因转染的人肝癌细胞系 2 2 15细胞培养中 ,对细胞的毒性及其分泌的表面抗原 (HBsAg)和e抗原 (HBeAg)的抑制作用。方法 (1)药物对细胞的毒性实验 ,在接种 2 2 15细胞后 2 4h ,加 2倍稀释的 6个稀释度药液 12 .0 0、8.0 0、4.0 0、2 .0 0、1.0 0、0 .5 0mg·ml-1,4d换一次药液 ,维持 12d ,用显微镜观察细胞病变 ;(2 )药效学实验 ,在无毒浓度 (4.0 0mg·ml-1)下 ,以 2倍稀释药液稀释为 4.0 0、2 .0 0、1.0 0、0 .5 0mg·ml-14个浓度 ,37℃ 5 %CO2 培养 ,每 4d收取培养液 ,换原浓度药液培养 ,并于第 12d时收集培养液 ,同时测定HBsAg和HBeAg。结果 (1)细胞毒实验结果表明 ,水芹提取物对细胞的半数细胞毒浓度 (TC50 )为 8.6 6 2± 0 .2 9mg·ml-1,最大无毒浓度 (TC0 )为 4.0 0mg·ml-1。 (2 )药效学试验 ,在最大无毒浓度4.0 0mg·ml-1对细胞分泌的HBsAg抑制率为 (6 9.1± 6 .6 ) % ,半数有效剂量 (IC50 )为 2 .15mg·ml-1;对细胞分泌的HBeAg的抑制率为 (78.9± 1.2 ) % ,IC50 为 1.0 4mg·ml-1。与细胞对照组比较均有非常显著性差异 (P <0 .0 1)。结论 水芹提取物在 2 2 15细胞中培养 12d ,无毒浓度对 2 2
Aim To study the inhibited effect of extract from Oenanthe Javanica on HBsAg and HBeAg in the cultured hepatocarcinoma cell line 2215(Hep G 2) which was genetransfered with hepatitis B virus(HBV) and its toxicity.Methods Twenty four h after 2215 cells vaccinated,the varied concentrations of Oenanthe Javanica added to cultured fluid,then exchanging the drugs every four days until to 12d.The cells alteration were observed under microcopy in order to evaluate the toxicity of Oenanthe Javanica.After treated with Oenanthe Javanica for 12d,the culture fluids were collected and the content of HBsAg and HBeAg were determined.Results The TC 50 of extract from Oenanthe Javanica were 8.662mg·ml -1 .The maximum nontoxic concentration (TC 0) were 4.00mg·ml -1 .In nontoxic concentration(4.00mg·ml -1 ),the inhibitions of Oenanthe Javanica on HBsAg and HBeAg were 69.1%(IC 50 =2.15mg·ml -1 ) and 78.9% (IC 50 =1.04mg·ml -1 )respectively(P<0.01;P<0.01).Conclusion The results show that Oenanthe Javanica can inhibite the secretion of HBsAg and HBeAg in cell line 2215 and no toxicity at high concentration.
