大鼠肾上腺髓质嗜铬细胞的原代培养及其儿茶酚胺的分泌 被引量:2

摘要 采用胶原酶消化,密度梯度离心及一次接种纯化等方法获得的大鼠肾上腺髓质嗜铬细胞(adrenalmedullarychromaaffincellsAMCC),原代培养可维持1月左右.在1周内细胞分泌功能稳定,对刺激剂反应良好. Isolated adrenal medullary chromaffin cells were Prepared by treatment of rat adrenalmedulla with collagenase collowed by purification with bovine serum albumin density-gradient centrifugationand differential Plating and were maintained in Dulbecco's Modified Eagle's Medium supplemented with 15 %calf serum. The primary cultUres could be maintsed for 4 weeks, but their secretory activity decreased after oneweek. In the first 6 days, the cells responded to secretagogus excellently.
