摘要
目的 研究大鼠肾上腺髓质嗜铬细胞钙离子通道的亚型。方法 利用全细胞膜片钳技术 ,在维持电压 - 70m V的条件下 ,给予细胞 4 0 ms,从 - 6 0 m V到 +6 0 m V的序列除级方波 ,阶跃 10 m V,引导出钙通道电流 (ICa) ,观察不同类型钙通道阻滞剂对 ICa的影响。结果 用 10 μmol/ L 硝苯地平 (NIF)、 1μm ol/ Lω- conotoxin GVIA和 10 0 nmol/Lω- agatoxin IVA灌洗细胞 10 m in,对 ICa有明显的抑制作用 ,抑制率分别为 (44 .6± 2 .0 ) %、 (31.3± 2 .0 ) %和 (2 0 .6± 2 .0 ) %。结论 在大鼠肾上腺髓质嗜铬细胞中 ,L 型钙通道是主要的 ,同时也存在 N型和 P/
Objective To investigate the subtypes of Ca 2+ channel in rat adrenal chromaffin cells. Methods By using the whole cell patch clamp technique, the effects of different kinds of Ca 2+ channel antagonists on I Ca by 40 ms depolaring pulse applied from a holding potential of -70 mV in 10 mV steps up to ±60 mV were observed. Results I Ca could be obviously inhibited by using 10 μmol/L nifedipine (NIF), 1 μmol/L ω conotoxin GVIA and 100 nmol/L ω agatoxin IVA to perfuse cells for 10 min. The inhibition rates of I Ca were (44 6±2 0) %, (31 3±2 0) % and (20 6±2 0) %, respectively. Conclusion In rat adrenal chromaffin cells L type Ca 2+ channels are predominant, but other subtypes of Ca 2+ channels like N type, P/Q type Ca 2+ channel are also present.
