摘要
目的:探讨糖皮质激素(GC)快速抑制肾上腺髓质嗜铬细胞(AMCC)受刺激后分泌的机制。方法:用Fura-2作Ca2+指示剂,观察GC对乙酰胆碱(ACh)刺激引起的AMCC内游离钙([Ca2+]i)浓度升高的影响,并用放射免疫方法测定了GC对ACh刺激后AMCC的环核苷酸含量的影响。结果:GC对ACh引起的AMCC的[Ca2+]i升高有快速抑制作用,但对ACh刺激引起的环核苷酸含量升高无明显作用。
Objective: Primary cultures of chromaffin cells from rat adrenal medulla were used to study the intracellular mechanism of the rapid inhibitory effects of glucocorticoids on the stimulated secretion. Methods: Loading chromaffin cells with Fura2, a fluorescent indicator of free cytosolic Ca2+ concentration ([Ca2+]i). [Ca2+]i were measures with fluorescence spectrometer. The effect of corticosterone and dexamethasone on cAMP and cGMP accumulation in chromafin cells stimulated by acetylcholine for 20 min was examined with radioimmunoassay. Results: The data showed that dexamethasone significantly inhibited the elevation of [Ca2+]i response to acetylcholine. The effect of corticosterone and dexamethasone on cAMP and cGMP was not evident. Conclusion: [Ca2+]i may play an important role in the mechanism of the rapid effects of glucocorticoids on the secretion of chromaffin cells.
