摘要
目的:探讨高效毛细管电泳法(HPCE)在测定桑叶中芦丁含量的作用。方法:选择新疆不同采集期桑叶。HPCE条件:熔融石英毛细管50cm×50μm,uncoated;硼砂和磷酸二氢钠缓冲溶液20mmol/L,其中含15%甲醇溶液,pH8.5;分离电压20kV(+)-(-);电泳时间12min;电泳温度25℃;压力进样0.5psi×10s;柱上检测UV254nm(二极管阵列检测器)。结果:样品可在10min内出峰,测得蒜氨酸浓度在O~80μg/ml范围内与峰面积的线性关系良好(r=0.9945),平均回收率为98.9%,批内RSD为3.9%。结论:高效毛细管电泳法准确、简单、快捷、重现性好。
Objective: To establish a HPCE method to determine the content of rutin in mulberry leaves. Methods: HPCE mettlod: bare fused-silica capillary 50 cm× 50 μm; borax buffer 20 mmol/L containing 15% methanol at pH8.5; running voltage 20 KV; polarity from anode (inlet) to cathode (outlet); separate time 12 mini temperature 25℃ ; sampling 0.5 psi×10 s; on line detection at 254 nm (PDA). Results: The sample could be separated in 10 min. The linearity relationship of rutin concentration and peak value was good in the range of 0-80 μg/ml. The related coefficient was r =0. 9945, the average recovery of sample for alliin was 98.9%, and the RSD of inter-day was 3.9%. Conclusion: This methods is simple, quick, accurate and with satisfactory recoveries.
