摘要
目的:建立侧柏叶中槲皮苷和杨梅苷的HPLC含量测定方法。通过对不同采集期和不同产地的侧柏叶中杨梅苷和槲皮苷含量变化的考察,为进一步控制侧柏质量提供数据基础。方法:采用Inertsil ODS-3(4.6 mm×250 mm,5μm)色谱柱,流动相为甲醇-0.01 mol·L-1磷酸二氢钾溶液-冰醋酸(37:63:1.5),流速1.0 mL·min-1,检测波长254 nm,柱温40℃。结果:杨梅苷与槲皮苷的色谱峰达到良好分离效果,线性范围分别为0.023~0.414μg(r=0.9999)和0.094~0.846μg(r=0.9999),平均加样回收率(n=5)分别为98.5%(RSD=0.42%)和101.3%(RSD=0.78%)。结论:本文所建立的侧柏叶HPLC含量测定方法简便快捷,精密度高,结果准确,为侧柏叶的质量控制提供了重要依据。
Objective:To establish a method for determination of myricetin and quercetin in Platycladus orientalis ( L. ) Franco and provide data basis for its further quality control through investigation of the content changes of myr- icetin and quercetin in the crude drugs from different collecting periods and different producing areas. Methods:The separation was performed on an Inertsil - ODS -3 (4. 6 mm × 250 mm, 5 μm)column, the mobile phase consisted of methanol -0. 01 mol · L-1 potassium dihydrogen phosphate solution- glacial acetic acid(37: 63: 1.5) ,with a flow rate of 1.0 mL · min - 1 at 40 ℃, the wavelength for measurement was at 254 nm. Results: A good linear relationship between the peak area and the mass existed in the ranges of 0. 023 - 0. 414 μg ( r = 0. 9999 ) for myricetrin and 0. 094 - 0. 846 μg ( r = 0. 9999 ) for quercitrin. The average recoveries ( n = 5 ) of myricetrin and quercitrin were 98.5% ( RSD = 0. 42% ) and 101.3% ( RSD = 0.78% ), respectively. Conclusion: The method is simple, rapid and accurate and can be used for quality control of Platycladus orientalis.
