Study on Characteristics of Xylanase from Trametes trogii

摘要 从威海市玛珈山上获得毛栓菌(Trametes trogii),在对其产酶条件研究的基础上,主要研究pH、温度、无机离子对毛栓菌木聚糖酶活性和稳定性的影响,以及该酶底物浓度效应和Km值测定。结果表明:在pH 6.81左右酶活性最高,pH在5.60-9.18范围内体现较强的稳定性.最适酶解温度为40℃,酶液在50℃以下有较好的热稳定性,Na^+、Mn^2+对木聚糖酶有激活作用,而Cu^2+、Fe^3+、Ca^2+则抑制木聚糖酶活性,表观Km值为1.25×10^-2g·L^-1。 Trametes trogii was screened from Majia Hill in Weihai. The effect of pH, temperature and inorganic ion on Trametes trogii xylanase activity and stability was studied. Meanwhile, the substrate concentration effect of this xylanase was also studied and Km was measured. The results showed:The optimum pH of the xylanase was 6.81,and the xylanase was stable when pH from 5.6 to 9.18. The optimum of xylanase was 40℃ ,and the xylanase could work continuously under 50℃. Na^+ , Mn^2+ had activation but Cu^2+ ,Fe^3+ ,Ca^2+ had inhibition on activity of xylanase. The Km of the xylanase measured 1.25 ×10^-2g·L^-1.
