摘要
从土壤中分离出一株产普鲁兰酶活性较高的菌株Z1,初步鉴定为盐球菌(Halococcussp.)。研究了其产酶最适条件,确定产酶最适碳源为糯米淀粉、最适氮源为酵母膏+蛋白胨,优化的培养条件如下:发酵初始pH值为6.0,接种量为14%,500 mL摇瓶装液量为150 mL,转速为180 r.min-1,培养温度为30℃,培养时间为52 h。在优化条件下产酶量达到最高峰,酶活力可达5.06 U.mL-1。
A bacterium producing pullulanase Z1 was screened from soil and identified to be Halococcus sp.. The optimum conditions for the pullulanase production were studied. The most suitable carbon source was sticky rice amylopectin and nitrogen source was yeast extract combined with peptone. The optimal fermentation conditions were determined as follows:initial pH value of 6.0,14% (volume fraction) amount of inoculation, 500 mL flask containing 150 raL liquid, 30℃ ,shaking at 180 r · min^-1 for 52 h. The enzyme activity of 5.06 U · mL^-1 was obtained under above conditions.
