摘要
采用高效毛细管电泳法对叶下珠中芦丁的含量进行测定。研究了缓冲液的种类、浓度、pH、电泳运行电压、进样时间等因素对分离检测的影响。结果表明,以60 mmol硼酸盐(pH 9.0)为缓冲液,16 mmol/L的SDS为添加剂,检测波长为245 nm,分离电压为15 kV,进样时间15 s,芦丁在20~500 mg/L线性关系良好(r=0.998),回收率为98.9%,RSD为2.1%,该法操作简单,回收率及重现性良好。
A method of high performance capillary electrophoresis was established for the detection of rutin in phyllanthus urinaria L. Effects of several important factors such as the pH and eoncentration of running buffer, concentration of SDS, operating voltage, and injection time, were investigated. Under the optimized condition using 60 mmol/L of Na2BO4 + 15 mmol of SDS (pH 9.0) as the running buffer at an operating voltage of 15 kV and a detection wavelength of 245 nm,the concentration of rutin demonstrated good linear relationship with the peak height in the range of 20 - 500 mg/L ( r = 0.998 9), with the average recovery rate reaching 98.9% and RSD equating to 2.1%.
