摘要
目的:研究大孔吸附树脂分离纯化玫瑰花中原花青素的最优工艺条件。方法:以静态饱和吸附量和解析率为指标,对五种大孔吸附树脂(D101、D1300、NKA、AB-8、NKA-Ⅱ)进行筛选,并以原花青素纯度为指标,采用正交试验法,分别考察了上样浓度、pH、洗脱液浓度和洗脱流速对纯化效果的影响。结果:D101型树脂综合性能最佳,适合纯化玫瑰花中原花青素,最优工艺条件为:上样浓度为1.25 mg/ml,上样pH为2,洗脱液为70%乙醇水溶液,洗脱流速为2 ml/min。结论:本研究可为分离纯化玫瑰花中原花青素提供方法。
Objective: To study the separation and purification technology for proanthocyanidins from rose and establish the best operating conditions. Methods : Evaluated by static adsorption capacity and elution ratio, five types of macroporous resins including D101, D1300, NKA, AB-8, NKA-Ⅱ were tested to separate and purify proanthocyanidins. And evaluated by product purity, the concentration of extract sample, pH of extract sample, concentration of eluant and flow rate of elution had been investigated. Results: D101 type macroporous resin showed the best property and was suitable for purifying proanthocyanidins from rose. The best operating conditions were as follows: 1.25 mg/ml as the concentration of extract sample, 2 as the pH value of extract sample, 70% ethanol equeous solution as the eluant and 2 ml/min as the flow rate of elution. Conclusion: This study can supply a method to separate and purify proanthocyanidins from rose.
