铅对原代培养小鼠肝细胞胆固醇7α-羟化酶基因表达影响 被引量:2

摘要 目的探讨醋酸铅对培养的小鼠肝细胞中胆固醇7α-羟化酶基因表达的作用,为解释铅与血脂之间的关系提供理论依据。方法提取20只昆明小鼠的肝细胞,进行离体培养,以不同剂量的醋酸铅作为处理因素,利用逆转录聚合酶链反应(RT-PCR)技术检测胆固醇7α-羟化酶基因的mRNA表达,并进行比较。结果不同剂量的醋酸铅对胆固醇7α-羟化酶基因转录水平的影响不同。高、中剂量组与对照组的差异均具有显著性(P<0.05或P<0.01),表现为随着染毒剂量的增加,胆固醇7α-羟化酶的mRNA表达量逐渐下降。结论醋酸铅作为一种暴露因素对胆固醇7α-羟化酶基因的表达有抑制作用。 Objective To study the direct action of lead on the expression of CYP7A1 gene which provides some information to explain the relationship between lead and blood lipid. Methods Firstly bepatocytes were extracted from Kunming neonatal mice and cultured in vitro, then treated with lead acetate at the doses of 0,1,3,9 p.mol/L for 24 hours, respectively. RT-PCR technique was utilized to detect the expression of CYP7A1 mRNA and compared with the control group. Results Different doses of lead acetate had differe t influence on the expression of CYP7A1 gene mRNA. Significant differences were observed at high and middle dose groups compared with the control group( P 〈0. 05 or P 〈0. 01 ), The more lead exposures, the lower expressions of CYP7A1 mRNA, Conclusion The expression of CYP7A1 mRNA can be inhibited by lead ( P 〈 0. 01 ).
