摘要
实验采用微量细胞培养噻唑蓝显色法观察丝裂霉素C(MMC)、阿霉素(ADM)、羟基喜树碱(HCPT)和噻替派(TSPA)等药物联合应用对膀胱癌细胞株BIU-87的毒性作用。结果显示HCPT与MMC、ADM联合应用能取得较好疗效。连续用药组预期50%抑制率剂量(ID50)与实测ID50的比值分别为13.59、11.26;序贯用药组先用HCPT,后用MMC、ADM、TSPA等药物,抑制率明显大于相同剂量药物单用抑制率。联合用药能增强化疗药物对BIU-87细胞的毒性作用。
This study was carried out to evalute the interactions of mitomycin C (MMC), adriamycin (ADM), hydroxycamptothecine(HCPT) and thiotepa(TSPA) against a human bladder cancer cell line BIU-87 by using the MTT assay in vitro. Results showed that combination of HCPT and MMC or ADM had the better synergistic cytotoxicity. In groups with continuons drugs using, the ratios of the median lethal dose between expectancy and actual result were 13. 59、11.26.In groups with sequential drug using, when HCPT was used first, followed by MMC, ADM or TSPA, the inhibition rate was obviously higher than when a single drug in the same dose was used. However, combination chemotherapy can increase cytotoxicity against BIU-87 line.
