摘要
目的探讨神经保护肽慢病毒(rLent/NT4-NAP)感染老年痴呆大鼠鼻粘膜后分泌的NAP对老年性痴呆(AD)大鼠行为学及海马内NF-kBmRNA表达的影响。方法选择无菌级雄性Wister大鼠40只,随机分为对照组,假手术组,AD组,rLent/NT4-NAP组,每组10只。建立β-淀粉样蛋白(Aβ)大鼠海马注射的实验性AD大鼠动物模型[2],利用水迷宫试验检测大鼠记忆能力,利用原位杂交的方法观察大鼠海马内NF-kBmRNA的表达。结果与AD组相比,rLent/NT4-NAP组水迷宫试验逃避潜伏期明显缩短,错误次数显著降低(P<0.01);原位杂交结果:AD组可见较多的NF-kBmRNA阳性神经元,rLent/NT4-NAP组大鼠海马区NF-kBmRNA表达较AD组明显减少,但较对照组NF-kBmRNA表达增多。AD组的平均光密度及平均灰度值均较对照组,假手术组,rLent/NT4-NAP组有显著差异(P<0.01),而在其他3组之间无显著差异(见表2)。结论NAP对老年性痴呆大鼠海马神经元具有保护作用,进一步证实了神经细胞中NF-kB活化受到抑制是可以延缓神经细胞凋亡的发生。
Objective To investigate Influence of NF-kBmRNA Expressing and praxiology of Alzheimer's disease mice in the hippocampus by Lentivirus neuroprotective peptid. Methods male Wister mice were randomly assigned into sham operation grop with 10 mice, control group with 10 mice, AD grop with 10 mice, rLent/NT4-NAP grop with 10 mice. Beta-amyloid protein injection into mice hippocamptts were applied to make experimental mice model of AD. The ability of learning and memory was tested by the water maze, NF-kB expressings were objected by hybridization in situ. Results the escape eclipse period of rLent/NT4-NAP grop is less than that of AD/group( P 〈 0.01 ). Damage neuron of rLent/NT4-NAP grop are less than that of AD/group. Conclusion treament of NAP can protect neuron of hippocampus in the Alzheimer' s disease mice.
