海马注射β淀粉样蛋白对大鼠学习记忆及局部神经元的损伤作用 被引量:41

摘要 目的 探讨β淀粉样蛋白 (Aβ)海马内注射的神经毒性 ,并对应用此方法建立阿尔茨海默病 (AD)的动物模型进行初步评价。方法 应用凝聚态Aβ1 4 0 进行大鼠海马齿状回背侧细胞带微量注射 ,观察 2d、10d、30d后学习记忆和神经病理损伤情况。结果 Aβ1 4 0 注射后大鼠迷宫学习记忆能力明显受损 ,获得记忆的尝试次数约为正常对照组的 3倍。注射区Aβ沉积、神经元死亡、胶质细胞浸润 ,背侧细胞带受损长度约为生理盐水注射组的 4~ 5倍。结论 凝聚态Aβ1 4 0 海马注射具有明确的在体神经毒性作用 ,可较好地模拟AD行为和病理表现 ;作为AD模型之一 。 Objectives To investigate the neurotoxicity of beta amyloid (Aβ) consisting of senile plaques in Alzheimer disease (AD) in the animal model. Methods 1 μl (10 μg) incubated Aβ 1 40 was injected into the dorsal blade of the dentate gyrus in the right hippocampus of each rat. The performance of Y maze task and the neuropathological effects by Aβ 1 40 injection were studied in the rats. Results The capability of acquisition and retrial of Y maze were impaired significantly in Aβ 1 40 treated rats 2 d,10 d and 30 d after injection, but no differences between three timepoints. Aβ deposition, neurons death and glial reaction were found in or around the injection sites. Conclusions These results suggest that the aggregated Aβ 1 40 and its deposition in the hippocampus is related to the impairment of learning and memory and the neuronal degeneration in the rats which showed similar behavior and pathological characterizations of AD. This Aβ treated rats could be used as an animal model for AD to probe the neurotoxic mechanisms of Aβ and its treatments.
