神经保护肽对老年性痴呆模型大鼠病理学及海马NF-κB mRNA表达的影响 被引量:1

摘要 目的探讨神经保护肽(NAP)对老年性痴呆模型大鼠病理学及海马内核因子(NF)-κB mRNA表达的影响。方法Wistar雄性大鼠40只,随机分为对照组,假手术组,痴呆组,治疗组,每组10只。建立β-淀粉样蛋白(Aβ)大鼠海马注射的老年性痴呆大鼠动物模型,利用HE染色观察病理变化,利用原位杂交的方法观察大鼠海马内NF-κB mRNA的表达。结果HE染色观察病理变化:治疗组可看到部分神经细胞的坏死,形状不规则,核固缩,染色加深,与痴呆组相比,神经元损伤情况明显减轻。原位杂交结果:痴呆组可见较多的NF-κB mRNA阳性神经元,治疗组大鼠海马区NF-κB mRNA表达较痴呆组明显减少,但较对照组NF-κB mRNA表达增多。痴呆组的平均光密度及平均灰度值与对照组、治疗组有显著性差异(均P<0.01)。结论NAP对老年性痴呆大鼠海马神经元具有保护作用。 Objective To investigate the influence of NAP on the expression of NF-κB mRNA and pathology of hippocampus in senile dementia rats. Methods 40 male Wistar rats were randomly divided into control, sham, dementia and treatment groups, 10 rats in each group. The senile dementia rats model was established by beta-amyloid protein injected into hippoeampus. The expression of NF-κB mRNA was measured by hybridization in situ. Results The condition of neuron injuries in treatment group was decreased than that in dementia group. The NF-κB mRNA positive neuron in dementia group was much more, and the expression of NF-κB mRNA in treatment group was decreased than that in dementia group, but increased than that in control group. The gray level and OD value in dementia group were different from those of control and treatment groups( P 〈 0.01 ). Conclusions NAP has the effect of neuroprotection on the hippocampus in senile dementia rats.
