Effect of nuclear factor-kappa B on pulmonary oxygen toxicity in rats

摘要 目的探讨核因子(NF)-κB在大鼠肺型氧中毒发病中的作用。方法32只SD大鼠随机分为正常对照组及230 kPa高压氧暴露2、6、10h组。观察肺组织病理变化以及NF-κB和肿瘤坏死因子α(TNF-α)在肺组织中含量的变化。结果(1)病理检查发现肺组织在高压氧暴露6h组出现炎症改变,10h组肺损伤加重。(2)肺组织细胞核中NF-κB P65含量在高压氧暴露2h组中明显增高,6h组中达高峰,10h组中下降但仍高于正常对照组。(3)肺组织TNF-α mRNA表达及肺匀浆中TNF-α浓度在高压氧暴露6h组中明显增高,10h组中继续增高。结论高压氧可以通过活化NF-κB诱导TNF-α/高表达从而介导氧中毒的肺损伤。 Objective To investigate effects of nuclear factor-kappa B (NF-κB) on pulmonary, oxygen toxicity in rats. Methods Thirty-two SD rats were randomly divided into 4 groups: normal control group; 2, 6 and 10 h of 230 kPa hyperbaric oxygen (HBO) exposure groups. Pathological change, the contents of NF-κB and TNF-α in lung tissue were observed. Results (1) Inflammation of lung was found after 6 h of HBO oxygen exposure and aggravated after 10 h of HBO exposure. (2) The contents of NF-κB P65 in lung tissue nucleus increased significantly after 2 h and reached the peak value after 6 h of HBO exposure. It declined after 10 h of HBO exposure but was still higher than the content of normal control group. (3) The expression TNF-α mRNA in lung tissue and content of TNF-α in lung homogenate increased significantly after 6 h and continued to increase after 10 h of HBO exposure. Conclusions HBO may activate NF-κB to elevate expression of TNF-α, which may lead to oxygen toxicity lung injury.
