摘要
目的:通过检测非类固醇类抗炎药(NSAID)氮-2,环己氧-4,硝基苯-甲基磺胺(NS398)对前列腺癌细胞株DU-145中RECK基因表达的调控,探讨其可能的抗肿瘤机制。方法:应用不同浓度的NS398作用于前列腺癌细胞株DU-145,培养48h后收集细胞,抽取组织总RNA,检测RECK基因与基质金属蛋白酶2(MMP-2)的表达;抽取组织总蛋白,应用Western blot方法检测RECK蛋白的表达。结果:各治疗组RECK基因表达均明显升高,其中以100μmol/L的NS398组升高最明显(P<0.05),MMP-2的含量则明显升高。另外,各治疗组RECK蛋白的表达也明显升高。结论:NS-398有明显的诱导RECK基因表达的作用,是其可能的抗肿瘤作用机制。
Objective: To investigate the regulatory effect of the nonsteroidal anti-inflammation drug NS398 on the RECK gene in the prostate cancer cell line DU-145. Methods: DU-145 was cultured with different concentrations of NS398 for 48 hours, the mRNA level measured by RT-PCR and the protein level by Western blot. Results: There was a significant increase in the mRNA level of the RECK gene in all the concentration groups, particularly in the 100μmol/L group (P 〈0.05), and an obvious decrease in that of MMP-2. The expression of the RECK protein was also significantly higher in the treated goups than in the control (P 〈 0.05 ). Conclusion: NS398 induces the expression of the RECKgene, which might be the mechanism of its anti-tumor effect.
