摘要
目的:观察慢性低O2高CO2肺动脉高压大鼠肝脏组织超氧化物歧化酶(SOD)、丙二醛(MDA)与转化生长因子β1(TGF-β1)表达变化及红花注射液对其的影响。方法:制备慢性低O2高CO2肺动脉高压大鼠模型,观察肝组织病理学改变;测定肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)的含量;免疫组化技术检测TGF-β1在肝内的表达及定位;RT-PCR技术检测肝组织TGF-β1 mRNA表达量。结果:(1)光镜下慢性低O2高CO2组大鼠肝组织脂肪变性明显,红花组肝组织脂肪变性不明显;(2)慢性低O2高CO2组大鼠肝组织MDA较正常组显著升高,而SOD显著下降,红花组大鼠肝组织MDA较慢性低O2高CO2组显著降低,而SOD显著升高;(3)慢性低O2高CO2组大鼠肝组织TGF-β1及其mRNA表达显著高于正常组,红花组大鼠肝组织TGF-β1及其mRNA表达显著低于慢性低O2高CO2组。结论:慢性低O2高CO2肺动脉高压状态下大鼠肝脏组织脂质过氧化增加,TGF-β1及其mRNA表达增加,红花注射液能减轻慢性低O2高CO2大鼠肝脏组织脂质过氧化作用并减少TGF-β1基因的表达。
Objective: To explore the influence of chronic hypoxia and hypercapnia on superoxide dismutase, malondialdehyde and transforming growth factor-beta 1 ( TGF-β1 ) gene expression in rat liver and the protective effects of Safflower Injection. Methods . The animal model of chronic hypoxia and hypercapnia was induced in rats . Histological examination were made and the level of malondialdehyde (MDA)and superoxide dismutase (SOD) were tested in all samples. The expression of TGF-β1 in liver tissue were detected by immunohistochemical technique and RT-PCR. Results : ( 1 ) Obviously fatty change was seen under optical microscope in hypoxic hypercapnia rats and less fatty change in rats that under Safflower injection administrations. (2) The MDA level was significantly higher and the SOD level was markedly lower in hypexic hypercapnia rats than those in normal animals . In the Safflower Injection administrations groups , the MDA level was lower and the SOD level was higher. (3) The expression of TGF-β1 protein and gene was increased significantly in the hypoxic rats. In the Safflower injection administrations groups, the expression of TGF-I31 protein and gene was lower than those in hypoxic hypercapnia groups. Conclusion: Chronic hypoxia and hypercapnia increases lipid peroxidation and the expression of TGF-β1 in the rats liver. Safflower Injection has protective effects on lipid peroxidative injury and deceases the the expression of TGF-β1 gene.
