摘要
目的探讨红花对增生性瘢痕成纤维细胞(HSFB)增殖和胶原合成的影响及其意义。方法通过MTT比色试验和生长曲线测定法检测红花对体外培养的HSFB增殖活性的影响;3H-脯氨酸掺入法测定红花对细胞胶原蛋白合成的影响;用光镜和电镜观察药物作用后HSFB形态学和超微结构的改变。结果红花可明显抑制HSFB的增殖和胶原蛋白的合成,且在一定范围内具有时间一剂量依赖性,以8μg/ml浓度组第3天作用最为显著;HSFB的形态和超微结构呈抑制改变。结论红花具有抑制HSFB增殖和胶原合成的作用,为应用该药治疗瘢痕提供了实验依据。
Objective To study the significance and effect of Carthamus tinctorus on proliferation and collagen synthesis of hypertrophic scar fibroblasts (HSFB). Methods Fibroblasts derived from human hypertrophic scar were cultured in vitro, the influence of Carthamus tinctorus on cell proliferation and collagen synthesis of the fibroblasts were investigated by MTT colorimetric assay, cell growth curve and ^3H-proline uptake analysis. Light microscope (LM) and electron microscope (EM) were employed to study the morphology and ultrastructure changes in fibroblasts after the application of Carthamus tinctorus. Results In definite range, Carthamus tinctorus could markedly inhibit the proliferation and collagen synthesis of HSFB in a time-dose manner, especially treated with 8μg/ml concentration for 3 days. Carthamus tinctorus obviously restrained the morphology and ultrastructure of fibroblasts. Conclusion Carthamus tinctorus can significantly inhibit the proliferation and collagen synthesis of HSFB, which may provide some foundation of treating pathological scar.
