吖啶橙与ctDNA相互作用机理的光谱法研究 被引量:5

摘要 应用光谱法研究了三环杂芳香类染料吖啶橙(AO)与DNA的相互作用,探讨了以AO为探针与小牛胸腺DNA(ctDNA)的作用机理。在pH2.00的Tris-HCl缓冲溶液中,AO在DNA的表面聚集作用导致体系的共振光散射(RLS)增强,在339nm处出现了新增强的RLS峰;通过紫外-可见吸收光谱观察,当DNA与AO的质量比超过0.166后,AO与DNA的作用方式经历了由嵌入到聚集的改变;进一步研究用荧光法作Scatchard图得出AO与DNA的作用方式为嵌入和静电的混合作用模式:每个核苷酸上的成键位点数n和每一位点的结合常数K均随AO浓度改变而发生了变化。此外,还研究了阴离子表面活性剂的协同作用等。 The interaction of acridine orange (AO) and DNA were studied by spectroscopic analysis methods. The interactional mechanism between ct DNA and AO in pH 2.00 Tris-HCl buffer solution was investigated as well. A new resonance light scattering peak at 339nm was found because of AO's assembling on the surface of DNA. Through the UV-Vis spectroscopy,interaction mechanism of AO and ct DNA changed from embedment to assemblement when ratio of DNA to AO exceeded 0. 166. The results from Scatchard Curve show that AO bound to DNA by mixture modes: intercalative binding and electrostatic attraction(the found of n orK altered when the concentration of AO changed). Furthermore,the resonance light scattering (RLS) of AO-DNA was greatly enhanced by anionic surfactant (AS), which concludes that AS has a concerted influence on the binding interaction of AO with DNA.
