摘要
目的探讨静脉注射骨髓基质细胞治疗大鼠局灶性脑缺血后的bFGF表达。方法采用大鼠局灶性大脑中动脉缺血1h模型,观察第3、7、14天生理盐水组,正常对照组,股静脉注射骨髓基质细胞组神经功能的恢复。免疫组化测定脑缺血再灌注第3、7、14天bFGF蛋白表达。结果神经功能评分:生理盐水组和正常对照组相比,各时间点均无显著差异(P>0.05);治疗组与对照组相比,治疗组第14天大鼠神经功能缺损评分减低,并且有显著性差异(P<0.05)。免疫组化结果:在梗塞半球大量BrdU阳性细胞,对侧半球也可见少量BrdU阳性细胞。bFGF蛋白表达:生理盐水组和正常对照组之间,缺血各时间点的表达均无显著性差异(P>0.05),治疗组与对照组相比各个时间点的表达均显著增加(P<0.05)。结论静脉注射骨髓基质细胞治疗大鼠局灶脑缺血的机制可能与bFGF蛋白表达增加有关。
Objective To investigate expression of bFGF with bone marrow stromal cells injected after the MCAO for 1 hour in rat and study the protective effect of BMSCs against injury and the possible roles of in the expression of bFGF following ischemia in rat. Methods Temporary ischemia of the MCAO for 1 hour was induced by the suture occlusion technique. We detected the expression of the bFGF after 3,7, 14 days of cerebral ischemic reinfusion. Results Significant recovery of neurological function was found in rats treated with BMSCs. After induction BMSCs transplantation in MCAO + MSCs group neurological deficits alleviated significantly at 14 days compared not only with the other groups at the same time points with the same group at different time points. The amount of BrdU positive cells within damaged areas in auto transplantation groups were significantly larger than that in the symmetrical brain area. Expression of bFGF protein: no obvious difference between the saline group and nothing group,the expression was no strinking difference at each temporal point after lacking of blood(P〉 0.05). The expression of the BMSCs group at each temporal point increased significantly(P〈 0.05 ). Conclusion The increase of bFGF in the ischemic tissue plays an important role in neurological recover. It protects nerve cell and neurological cell and promote new vessel to form, therefore it protects its brain.
