摘要
目的:研究低剂量电离辐射对小鼠睾丸生精细胞中活性氧(ROS)活性和线粒体膜电位(△Ψm)的影响。方法:以2′,7′-二氢二氯荧光黄双乙酸钠(DCFH-DA)和罗丹明123(Rhodamine 123)为探针,采用流式细胞术,间接检测细胞内ROS活性和△Ψm。结果:不同剂量X-线照射后12h,小鼠睾丸生精细胞中ROS活性较0Gy组增加,并且随剂量增加而增加(P<0.05);△Ψm则表现为随剂量增加而降低(P<0.05)。0.075Gy X线照射后与0h比较,ROS活性随时间推移而增加(P<0.05),12h达峰值,并且维持在较高水平;△Ψm则随时间推移而降低(P<0.05),12h降到最低后逐渐恢复到正常水平。结论:低剂量电离辐射能够诱导小鼠睾丸生精细胞中ROS活性增加,而△Ψm降低,并且具有一定的剂量和时程效应规律性。
Objective To investigate the efftcets of low dose X-ray radiation on the activity of the reactive oxygen species (ROS) and mitochondrion membrane potential (ΔΨm) in spermatogenic cells of mouse testes. Methods The ROS activity and ΔΨm in the cells were measured indirectly by flow cytometry with 2', 7'- dichlorofluorescin diacetate (DCFH-DA) and Rhodamine 123 probes. Results The ROS activity increased with the increase of irradiation doses 12 h after irradiation with different doses of X-rays compared with 0 Gy (P〈0.05) ; but the ΔΨm decreased with the increase of irradiation doses compared with 0 Gy (P〈0.05). After irradiation with 0. 075 Gy X-rays, the ROS activity increased with time prolongation compared with 0 h (P〈0.05), reached the peak at 12 h later, and kept the higher level; but ΔΨm decreased with the time prolongation, compared with 0 h (P〈0.05), and reached the lowest level at 12 h later, then recovered to the normal level gradually. Conclusion Low dose radiation can induce the the increase of ROS and decrease of ΔΨm in spermatogenic cells of mouse testes, and has dose- and time-effect regulations.
