摘要
为了解贲门癌变的分子生物学发生基础,采用免疫组织化学方法,观察取自食管癌高发区河南林州市的37例贲门癌前病变组织P16和Rb肿瘤抑制基因蛋白的表达,并对二者的免疫组化结果进行比较。结果:贲门上皮各级病变组织均出现不同程度的细胞核P16和Rb蛋白免疫阳性反应。且随着贲门上皮病变的加重,P16蛋白免疫阳性率呈下降趋势,而Rb蛋白免疫阳性率呈升高趋势,两者具有显著的负相关性(P<0.05)。提示:P16和Rb蛋白的变化可能是贲门癌变早期的重要分子学改变。
To further understand the molecular basis of gastric cardia carcinogenesis, preliminary studies with immunohistochemistry method were carried out on the expression of tumor suppressor gene P16 and Rb in gastric cardia precancerous lesions from 37 subjects in high incidence area of gastric cardia cancer. The immunostaining results of the two parameters were compared. The present results demonstrated that the different degrees of nuclear immunostaining of P16 and Rb occurred on gastric cardia epithelium in the different stages of lesions. But, with the lesions progressing, the positive immunostaining rate of P16 protein had a decreasing tendency, and on the other hand, the positive immunostaining rate of Rb protein had a increasing tendency. Correlation analysis showed an significant negative relation between these two parameters (P<0.05). The present results indicated that the alteration of P16 and Rb protein was the important molecular events in the early stage of gastric cardia carcinogenesis.
