垂体腺瘤中半乳糖凝集素-3的表达及其与Bcl-2、Bax关系

Expression of galectin-3 in pituitary adenomas and its relationships with Bcl-2 and Bax

摘要 目的检测垂体腺瘤中半乳糖凝集素-3(Gal-3)、Bcl-2和BaxmRNA和蛋白的表达水平,探讨Gal-3和凋亡相关基因Bcl-2、Bax的内在联系。方法RT-PCR检测20例垂体腺瘤Gal-3、Bcl-2和Bax mRNA的表达;免疫组化染色检测78例垂体腺瘤中Gal-3、Bcl-2和Bax蛋白表达,分析侵袭性和非侵袭性垂体腺瘤中Gal-3、Bcl-2和Bax表达的差异及其相关关系。结果侵袭组垂体腺瘤Gal-3和Bcl-2mRNA和蛋白的表达水平较非侵袭组显著增高,而Bax mRNA和蛋白在两组中的表达水平差异无显著性意义。等级相关分析显示Gal-3蛋白与Bcl-2蛋白之间有正相关关系(r=0.343,P=0.002);Gal-3蛋白与Bax蛋白之间无相关关系(r=-0.170,P=0.136);Bcl-2蛋白与Bax蛋白表达之间呈负相关关系(r=-0.292,P=0.009)。结论Gal-3可能主要是通过与Bcl-2协同作用,发挥其抗凋亡效应,相对促进垂体腺瘤细胞增殖。 Objective To study mRNA and protein expressions ofgalectin-3 (Gal-3), Bcl-2 and Bax in pituitary adenomas, and explore the internal relations between Gal-3 and apoptosis-related genes, Bcl-2 and Bax. Methods RT-PCR was applied to detect mRNA expressions of Gal-3, Bcl-2 and Bax in 20 cases with pituitary adenomas; the immunohistochemical staining was applied to determine protein expressions of Gal-3, Bcl-2 and Bax in 78 cases with pituitary adenomas, analyzing the differences in expressions of Gal-3, Bcl-2 and Bax and their interrelations in pituitary adenomas of invasive and non-invasive groups. Results The mRNA and protein expression levels of Gal-3 and Bcl-2 in invasive pituitary adenoma group was remarkably higher than that in non-invasive group, but Bax mRNA and protein expressions did not differ significantly between the two groups. The rank correlation analysis showed that Gal-3 protein had positive linear correlation with Bcl-2 protein (r=0.343, P=0.002); Gal-3 protein had no linear correlation with Bax protein (r=-0.170, P=0.136); pearson correlation coefficient between Bcl-2 protein and Bax protein was r=-0.292, P=0.009 and they were negatively correlated with each other. Conclusion Gal-3 might exert its anti-apoptotic effect and promote cell proliferation of pituitary adenomas through its interaction with Bcl-2.
