摘要
目的探讨凋亡蛋白抑制因子生物素(Survivin)基因在脑胶质瘤中的表达与其在脑胶质瘤恶性增殖和抗细胞凋亡中的作用,及与Bcl-2基因在脑胶质瘤中的相关性。方法选择50例脑胶质瘤标本,采用免疫组化链霉卵白素生物素复合物(SABC)方法检测Survivin和增殖细胞核抗原(PCNA)蛋白及bcl-2的表达,分别计算Survivin免疫反应评分(IRS)和增殖指数(PI);采用TUNEL方法检测脑胶质瘤凋亡细胞,计算凋亡指数(AI)。结果脑胶质瘤Survivin IRS、PI及AI分别为3.8±3.9、15%和0.9%,三者均随脑胶质瘤病理级别的增高而升高(P均<0.05)。Survivin阳性组PI明显高于阴性组PI(P<0.05),PI与Survivin IRS呈显著正相关(r=-0.372,P<0.001)。Survivin阳性组与阴性组之间AI差异无统计学意义(P>0.05),但AI与Survivin IRS呈显著负相(r=-0.372,P<0.001)。结论 Survivin在脑胶质瘤中高表达,随病理级别增高而明显升高。其在脑胶质瘤恶性增殖和抗细胞凋亡过程中发挥重要作用。Bcl-2可以上调生物素的抗凋亡效应,两者呈正相关。
Objective to discuss the expression of apoptotic protein inhibitor gene, surviving, in the gliomas of brain and its function in the proliferation of malignant glioma and anti-apoptotic effects and its correlations to Bcl-2 gene in glioma. Methods In 50 specimen of glioma collected, we use SABC to monitor the expression of survivin, PCNA and bcl-2, analyzing the IRS and PI of surviving; apply TUNEL to monitor the apoptotic cell in glioma to get AI. Results the IRS, PI and AI of glioma are 3.8 ± 3.9,15 % and 0.9 % respectively, all the indexes increase when the pathological grade of glioma elevate ( P 〈 0.05 ). the PI in surviving positive group is significantly higher than the negative group( P 〈 0.05), PI and IRS of surviving is positively correlated ( r = - 0. 372, P 〈 0. 001 ). The AI of surviving positive group and negative group are not stastically related ( P 〉 0.05), but AI is negatively correlated with the IRS of survivin ( r = - 0. 372, P 〈 0. 001 ). Conclusion survivin is highly expressed in glioma and increases when the pathological grade elevates. It plays an important role in glioma malignant proliferation and anti-appoptotic process. Bcl-2 can up-regulate'the anti-apoptotie effects of survivin, they had a positive relationship.
