摘要
从激活的人T淋巴细胞λgt10cDNA文库中筛选含免疫反应性生长激素cDNA序列的克隆,并分析其核苷酸序列。结果表明人淋巴细胞免疫反应性生长激素全部阅读框架的cDNA序列与人垂体生长激素的完全相同。在基因水平证实了神经内分泌与免疫系统共用信息传递因子的理论。
Immunoreactive growth hormone (irGH) has been found to be expressed in human lymphocytes at low level, and has been shown to be similar to pituitary GH in terms of antigenicity, bioactivity, molecular weight and the mRNA size. Until now nucleotide sequence encoding irGH has not been reported. In the present study, 12 positive phage clones containing irGH cDNA by screening a λ gt10 cDNA library of activated human T lymphocytes were isoeated by using a pituitary hGH cDNA probe. The open reading frame (ORF) of lymphocytic irGH cDNA was amplified by PCR with specific primers from 8 of the 12 positive clones and 3 were subcloned into pUC12 plasmid vector. Dideoxy mediated chain termination DNA sequencing of these cDNA reveals that the sequence of irGH cDNA is identical to that of the pituitary hGH cDNA, and the ORF sequences of GH from both lymphocyte and pituitary are of 651 base pairs encoding same signal peptide of 26aa and mature hGH peptide of 191aa. The sequence of total ORF of human lymphocyte irGH cDNA is reported for the first time.
