芪术合剂干预肺纤维化大鼠TGF-β_1 mRNA表达的实验研究 被引量:3

摘要 目的:观察中药芪术合剂对博莱霉素致大鼠肺间质纤维化的干预作用及其机制。方法:将60只健康SD大鼠随机分为正常组、模型组、芪术合剂组和强的松组。经气管滴入博莱霉素复制肺纤维化大鼠模型,分组予以治疗,于治疗3、7、21 d分批处死各组大鼠,每次每组6只。观察肺组织病理变化和TGF-β1 mRNA的表达。结果:模型组在不同时段TGF-β1 mRNA表达较正常对照组明显增高,芪术合剂组与同期模型组比较明显降低,芪术合剂组和强的松组经干预后肺泡炎和肺纤维化有所减轻。结论:中药芪术合剂具有显著抑制博莱霉素致大鼠肺纤维化的作用,抑制TGF-β1 mRNA的表达是其可能的机制之一。 OBJECT Observe the effect and mechanism on bleomycin - induced pulmonary fibrosis in rats interfered with QiZhuHeJi. METHOD 60 SD rats were divided into four groups randomly: normal control group, model group treated with bleomycin, QZHJ treated group, and hydrocortisone treated group. The rats were duplicated pulmonary fibrosis model by intratracheal instillation of bleomycin, and each group was given corresponding drugs respectively. The experimental rats were sacrificed in batch on the 3rd ,7th and 21st day for observing TGF - β1 mRNA expression and pathological change. RESULT TGF - β1 mRNA expression in different stage of model group increased obviously and showed significant diffence in comparison with normal group. TGF- β1 mRNA expression in QZHJ treated group decreased significantly compared with hydrocortisone treated group in the same stage. And there were distinct differences. After intervention, airsacculitis and fibrosis were improved in QZHJ treated group and hydrocortisone treated group. CONCLUSION QZHJ has a notable effect in inhibiting rats pulmonary fibrosis induced by bleomycin, and TGF- β1 mRNA expression inhibited may be one of its mechanisms.
