摘要
目的:观察第三代维A酸药物芳维A酸氨丁三醇(代号FY-10)对人角质形成细胞环氧化酶-2表达的影响。方法:通过体外培养HaCaT细胞,脂多糖(LPS)诱导环氧化酶-2(COX-2)mRNA表达,再以芳维A酸氨丁三醇处理细胞后,采用RT-PCR的方法检测COX-2表达水平的变化。结果:(1)以10μg/L终浓度LPS作用细胞2h后,细胞COX-2 mRNA表达明显高于作用前(x-±s分别为0.779±0.099和1.033±0.207,t值为6.083,P=0.00012)。(2)在加入LPS的同时加入芳维A酸氨丁三醇可以明显降低COX-2 mRNA的表达,二者的x-±s分别为1.161±0.130和0.849±0.056,(t值为5.398,P<0.01)。(3)芳维A酸氨丁三醇的作用呈浓度依赖性,以1×10-6mol/L作用最强。结论:芳维A酸氨丁三醇对人角质形成细胞的环氧化酶-2有肯定的抑制作用,为其治疗以炎症细胞浸润为表现的银屑病提供了理论基础。
Objective: To detennine the effect of the retinoic acid (FY- 10) on the expression of cyclooxygenase 2 (COX- 2) of human keratinocytes (HaCaT cell line). Methods: Lipopolysaccharide(LPS) inducing COX- 2 mRNA expression in vitro cultured human epidermal keratinocyte was determined by reverse transcriptase polymerase chain reaction (RT- PCR) before and after treatment with FY- 10. Results: (1)The expression of COX- 2 was significantly increased after HaCaT cell was treated with LPS 10 ug/L final concentration for 2 hours or longer. (2) FY- 10 distinctly depressed the expression of COX- 2 induced by LPS. (3)The effect of FY- 10 was concentrat ion dependent, which was strongest in a concentration of 1 × 10^-6 mol/L. Conclusion: FY- 10 can definitely depress the expression of cyclooxygenase 2 in HaCaT cell line, which provides a base for the treatment of psoriasis characterized by infiltration of inflammatory cells.
